中文摘要 | 本研究探討二氧化硫及pH值對蘋果酸乳酸菌分解L-蘋果酸之影響。所採 用之蘋果酸乳酸菌包括Leuconostoc oenos PSU-1.CSb5及一株同型乳酸醱酵菌BB菌 株。菌體濃度及對L-蘋果酸分解能力之線性關係依菌株而異。L. oenos CSb5對二 氧化硫最為敏感;然而其貝有最高之L-蘋酸分解能力。L-蘋果酸分解之最佳pH值 因菌株而異;對L. oenos PSU-1,CSb5及同型乳酸醱酵菌BB而言,其最佳L-蘋果酸分 解之pH值分別為4.0-5.0,4.5及4.0。雖然L-蘋果酸無法作為蘋果酸乳酸菌之能量來 源,然其卻有提高該菌此生長速率之效果,且此一作用非來自於L-蘋果酸分解所 導致pH值上升之結果。由菌體內L-蘋果酸分解試驗顯示,二氧化硫對L-蘋果酸分 解抑制之作用機制可能為分子態二氧化硫和酵素結合或來自於二氧化硫之非競 爭型抑制作用。 |
英文摘要 | The effect of sulfur dioxide and pH on the L-malate degradation by malolactic bacteriawas examined. Malolactic bacteria, including Leuconostoc oenos PSU-1, CSb5, and a homolactic strain BB were grown in CBB medium at two pHs (pH 3.4 and 5.5). The linearrelationship between L-malic acid degradation and cell concentration was strain-depend.Leuconostoc oenos CSb5 was the most sensitive strain to sulfur dioxide; however, it showedthe highest L-malic acid degrading activity among the strains tested. The optimal pH for L-malic acid degradation varied between strains, for L. oenos PSU-I, CSb5 and the BBstrain, the optimal pH values were 4.0 to 5.0, 4.5, and 4.0, respectively. Although L-malicacid is not an energy source for the malolactic organism, the specific growth rate of thesebacteria was found to be increased by the presence of malic acid. The stimulation ofgrowth did not result from changes in pH that accompany L-malate degradation. Themechanism of sulfur dioxide inhibition of malic acid degrading activity suggested from invivo studies indicates that malate degradation may be inactivated due to molecular sulfurdioxide binding of the enzyme or by noncompetitive inhibition of sulfur dioxide. |
