中文摘要 | 本實驗主要探討龍鬚菜Gracilaria coforvoides生成前列腺素E2(prostaglandin E2, PGE2)的最適生成條件,進而探討不同季節G. coforvoides中PGE2含量的變化並比較不同品種的龍鬚菜中PGE2的含量,其品種包括了G. coforvoides、G. gigas及一種未知品種的野生龍鬚菜。結果顯示龍鬚菜G. coforvoides生成PGE2的最適反應溫度為25℃,在25℃下反應PGE2的生成量約為35℃者的2.38倍。G. coforvoides生成PGE2的最適反應PH為8.5,龍鬚菜萃取液的原始pH約為5.75,在pH 8.5的條件下反應PGE2的生成量約為pH 5.75者的29倍。在季節的影響方面,G. coforvoides中PGE2的原始含量(未添加花生四烯酸)並無因季節的不同而產生規則性的變化,但G. coforvoides萃取液加入花生四烯酸後PGE2的生成量會受季節的影響,在12~5月份這段期間PGE2的生成量較高,而在夏季時期PGE2的生成量較少尤其在7月份時PGE2的含量最低。不同品種的龍鬚菜PGE2的含量也有差異,其中以G. coforvoides PGE2的含量最高。因此龍鬚菜中PGE2的含量受生產條件(溫度、pH和花生四烯酸之添加)和品種而影響。 |
英文摘要 | The purpose of this study is to investigate the optimal conditions of prostaglandin E2 (PGE2) production and seasonal variation of PGE2 level in Gracilaria coforvoides. Furthermore PGE2 level in different species of Gracilaria was also analyzed involving G. coforvoides, G. gigas and another species Gracilaria sp. The result showed the optimal temperature of PGE2 production in G. coforvoides was 25℃. The PGE2 level at 25℃ was 2.38 fold as that at 35℃. The optimal pH of PGE2 production in G. coforvoides was 8.5. PGE2 level in pH 8.5 was 29 fold as that of the original pH of extract solution (pH 5.75). The PGE2 levels (without adding arachidonic acid) in G. coforvoides were not shown regular seasonal variation. Otherwise there were higher PGE2 levels (with adding arachidonic acid) in December-May than in summer season. Particularly, there was the lowest PGE2 level in July. The different PGE2 levels in different Gradilaria species were found. PGE2 level in G. coforvoides was the highest among tested three species. Therefore PGE2 levels in Gradilaria are different depending on forming factors (temperature, pH and adding of arachidonic acid) and species. |
