中文摘要 | 神經管是由胚胎的外胚層經神經板、神經溝、神經皺襞等變化最後成對的神經皺襞併合形成。神經管閉合不全常導致各種不同型式中樞神經系統先天性畸形,目前對其形成的病因仍然有爭論,為了要確實瞭解其形成的病因必須先瞭解正常神經孔閉合的過程。 本實驗是以掃描式電子顯微鏡及光學顯微鏡觀察大自鼠胚胎神經孔閉合的過程。大白鼠胚胎在第11天時其中段部已併合形成神經管,在第11.5天時大部份神經管已形成僅前、後神經孔仍開放著,在第12天時前神經孔已閉合僅後神經孔開放著,在第12.5天時後神經孔閉合神經管完全形成。以掃描式電子顯微鏡觀察未併合神經皺襞的表皮細胞與神經細胞交接處發現許多有疱狀突出的細胞,此細胞的疱狀突出兩側神經皺襞併合繭的轉變,以利神經皺襞早期的黏著。在組織切片檢查發現前神經孔的閉合是神經皺襞的表皮細胞先接合然後神經組織再併合,而後神經孔的閉合神經組織與表皮細胞同時接合。此結果來探討由ETU誘發大白鼠胎兒有Lumbosacral Myeloschisis with Hindbrain crowding畸型形成的肩因,可得一部份解釋。 |
英文摘要 | Neuropore closure was studied in rat embryos by light and scanning electron microscopy (SEM). The so-called neural folds are complex structures containing ectoderm (extemal surface), neuroectodenn (intemal surface) and, in between, an altered zone which is considered a prerequisite for adhesion between neural folds. The anterior neuropore closed on day l2 of gestation. Fusion of folds in this region was initiated by contact between the surface ectcderm and then neural tissue. The posterior neuropore closed on 12.5 of gestation; fusion in this region was accomplished by contact simultaneously between the neural tissue and surface ectodenn. The results of the present study may explain partly the pathomechanism of lumbosacral myeloschisis associated with hindbrain crowding induced by ethylenethiourea (ETU). |
