中文摘要 | 病毒誘導基因靜默(virus-induced gene silencing, VIGS) 是利用農桿菌攜帶含有目標基因片段的病毒載體, 農桿菌感染植株後干擾目標基因 RNA 量以抑制目標基因表現的技術,常應用於植物基因功能的研究。不同的農桿菌 系種類可能會影響基因靜默效果與距離,為了瞭解農桿菌系對基因靜默效果的空間性與時序性之影響,本實驗利用 LBA4404 與 GV3101 兩種農桿菌系,攜帶含有八氫番茄紅素合酶 (phytoene desaturase, PDS) 基因的菸草脆裂病 毒 (tobacco rattle virus, TRV) 載體,接踵於菸草 ( Nicotiana tabacum L. cv Havana) 子葉。發現 LBA4404 的 VIGS 效果較強但時效性短,而 GV3101 的傳播力較佳且效果較持久。本實驗之結果將有助於更精準的利用 VIGS 技 術應用於植物基因功能之研究。 |
英文摘要 | Virus-induced gene silencing (VIGS) is a technique that uses Agrobacterium to carry a viral vector containing a fragment of the target gene. After Agrobacterium infects the plant, it interferes with the RNA levels of the target gene to suppress its expression. This method is commonly used in the study of plant gene functions. Different Agrobacterium strains may affect the efficiency and distance of gene silencing. To understand the spatial and temporal effects of different Agrobacterium strains on gene silencing, this experiment utilized the Agrobacterium strains LBA4404 and GV3101. These strains carried a tobacco rattle virus (TRV) vector containing the phytoene desaturase (PDS) gene, which was inoculated into the cotyledons of tobacco (Nicotiana tabacum L. cv Havana). The results demonstrated that LBA4404 exhibited a stronger but shorter-lived VIGS effect, while GV3101 had better propagation and a more sustained effect. The results of this experiment will contribute to a more precise application of VIGS technology in the study of plant gene functions. |
