中文摘要 | 牛樟由於族群數量越來越稀少,致使有效種子不易產生,使得牛樟瀕臨絕種邊緣;後由於扦插技術提升,扦插苗成為牛樟造林苗木的主要來源。因此採穗園的建立格外重要,目前採穗園牛樟的根系中有哪些可培養的根部內生菌尚未釐清。本研究利用南投魚池牛樟採穗園之牛樟樹為試驗材料,利用光學顯微鏡進行染根觀察,可於根菌結合體內發現暗色隔膜菌絲及微菌核等構造;並於根系中共計誘導純化出132株菌株,依菌落顏色、大小及型態共計分為15類群;再依致病性結果得知有6株菌株對油菜不會產生致病性;經分子生物分析得知,CkYCP 1、CkYCP 11、CkYCP 12菌株屬於不完全菌類,CkYCP 4和CkYCP 14菌株屬於子囊菌門,CkYCP 2菌株屬於擔子菌門。綜合上述,本研究證實,南投魚池牛樟採穗園之根菌結合體具有暗色隔膜內生菌及叢枝菌根之菌根形態,並可從根系結合體中分離純化出6株不具致病性菌株。 |
英文摘要 | Cinnamomum kanehirae seeds have an entomophilous characteristic and can't be easily collected. Therefore, using cutting to propagate is the best silvicultural practice for the resources conservation of C. kanehirae. It is extremely important to set up a scion garden for propagation of C. kanehirae. At the moment, few researches have been conducted on cultivable root endophytic fungi from a scion garden of C. kanehirae. In this study, the root-fungus associations were collected from two scion gardens of C. kanehirae in Nantou Yuchih area. The like-vescular and hyaline microsclerotia were observed in root cells by light microscopy. One hundred thirty-two endophytes were isolated from C. kanehirae roots. These isolates were divided into 15 groups by morphology and characteristics. Among these 15 groups, six of them successfully passed pathogenicity testing. Molecular analysis indicated strains CkYCP 1, CkYCP 11 and CkYCP 12 belonged to Fungi imperfecti, and CkYCP 4 and CkYCP 14 belonged to Ascomycota, and strain CkYCP 2 belonged to Basidiomycota. This study demonstrated that C. kanehirae from the two scion gardens in Nantou Yuchih area could form arbuscular mycorrhiza and dark septate endophyte in the field. Thus, these six strains belonged to nonpathogenic fungi. |
