中文摘要 | 本研究探討補充葉酸對慢性腸炎誘導惡性結腸腫瘤進展之影響與調節惡性轉移相關之癌幹細胞分子指標。將3週齡雄性C57BL/6小鼠分為對照組、短期葉酸補充(STF)組與長期葉酸補充組(LTF)(3 mg/dL葉酸飲水),餵食三週後,腹腔注射Azoxymethane(AOM)(7.5 mg/kg body wt),一週後以2.5% Dextran sulfate sodium salt(DSS)飲水餵食5天。短期葉酸補充於AOM/DSS處理後1個月犧牲,長期葉酸補充組於AOM/DSS處理後5個月犧牲。結果發現短期葉酸補充組的總體腸炎活性評估指標(1.72)低於對照組(3.25)。相較於對照組,葉酸補充組在中段結腸部位惡性腫瘤發生率為75%,且無潰瘍產生。短期與長期葉酸補充組則有減少30-50%腫瘤顆數(P = 0.086)與顯著降低大腫瘤的數目(P = 0.017)。由western blotting可知E-cadherin與Gli1蛋白表現量在LTF組顯著低於STF組。OCT4蛋白表現量在LTF組顯著高於C組。綜上所述,長期葉酸補充顯著緩解炎症、結腸炎、潰瘍性病變和中段結腸惡性腫瘤發生率,且有降低轉移訊息轉錄子Gli1表現並促進胚胎幹細胞印記表現。 |
英文摘要 | This study investigated the effects of folate supplementation on the progression of chronic enteritis-induced malignant colon tumors and the molecular markers of cancer stem cells associated with malignant metastasis. Three-week-old male C57BL/6 mice were divided into control group (C), short-term folate supplementation group (STF) and long-term folate supplementation group (LTF)(3 mg/dL folic acid drinking water). Mice were intraperitoneal injection of Azoxymethane (AOM) (7.5 mg/kg body weight) after feeding 3 weeks. One week later, mice were fed with 2.5% Dextran sulfate sodium salt (DSS) drinking water for 5 days. DSS treatment was repeated three times to induce a chronic enteritis colon cancer model. The results showed that DAI in the LTF group was lower than C group when the third DSS treatment. Tumor tissue sections demonstrated that the incidence of malignant tumors was 100% in the intestines in the control group and was 75% in the middle segment of colon in the folate supplementation group and there was no ulcer. There was a 30-50% decrease in tumor numbers and a significant reduction in the large tumor numbers in the STF and LTF group. The protein expression of E-cadherin and Gli1 were significantly lower in the LTF group than STF group. OCT4 protein expression was significantly higher in the LTF group than C group. In summary, long-term folate supplementation significantly alleviated inflammation, colitis, ulcerative lesions and the incidence of colonic malignancies in the middle segment, and reduced the expression of the transcriptional message transcript Gli1 and promoted embryonic stem cell imprinting. |
