中文摘要 | 石斛蘭具有觀賞及藥用價值,然而無性繁殖速度慢,有必要建立種苗量產繁殖技術。本研究以組織培養之春石斛蘭苗為材料,取其假球莖上的休眠芽離體組織培養,探討暗培養及刻傷處理對芽體形態發生之影響。結果顯示,自Dendrobium Red Emperor 瓶苗取芽體培養,未經暗處理之芽體,施用1 mg.L^(-1) 1-naphthalene acetic acid (NAA)對芽體形成營養芽有促進之效果,但培養10 週以上芽體表現逐漸衰弱,高濃度NAA 甚至會抑制植株之形成;而6 週暗處理則不利於芽體分化。芽體在培養初期以含馬鈴薯及生長調節劑的培養基培養,存活率較高,另外配合刻傷處理及固、液體交互培養,可誘導長出單芽、雙芽及多芽等不同形態,培養12 週後由芽體形成植株之增殖倍率為1~3 倍。 |
英文摘要 | Dendrobium has ornamental and medicinal values, however its asexual propagation rate by division is slow. It is necessary to establish a rapid seedling production method. In this study, we used bud explants of in vitro cultured seedling as materials, investigated the effects of dark culture and different NAA concentrations, medium and wounding treatment on the growth of bud explants of nobile-type dendrobium orchid. Results showed that in normal light culture, NAA 1 mg.L^(-1) could promote the growth of bud explants but then became weaker after 10 weeks of culture. Higher NAA concentration could inhibit bud explants formation of shoots. Dark culture caused adverse effects on bud growth and development. At the initial stage of culture, medium contain potato and plant growth regulator had higher bud survival rate. Wounding treatment in conjunction with solid-liquid culture could induce multiple shoot growth. Starting from a bud, the proliferation rate is 1~3 plantlets after 12 weeks of culture. |
