中文摘要 | 由於關節炎等骨科相關疾病與體內自由基的增加有關,本實驗檢測台灣民間常用食補骨科草藥牛乳房的抗氧化能力,以評估其生理功效。牛乳房分別以70%的酒精浸泡或熱水熬煮以進行萃取;之後以清除DPPH自由基能力、清除超氧化物(superoxide)能力、抑制脂質過氧化力及DNA保護能力等檢測其抗氧化力,並分析萃取物中總酚類物質的含量。結果顯示,牛乳房酒精萃取物和水萃取物清除DPPH自由基(IC(下標 50)分別為:2.65 mg/mL及36.66 mg/mL)和超氧化物自由基(IC(下標 50)分別為3.71 mg/mL及>5 mg/mL)的能力也較catechin差,但卻能較catechin有效地抑制大鼠肝臟脂質過氧化(p<0.05)。此外,牛乳房也能減少ΦXl74 supercoiled DNA受到由UV light和H2O2所帶來的傷害。在分析總酚物質含量時,顯示牛乳房酒精萃取物的總酚含量為水萃取物的二倍,此一總酚含量差異與酒精及水萃取物的抗氧化能力差異一致。 |
英文摘要 | Increased concentration of free radicals in vivo contributes to the etiology of musculoskeletal diseases such as arthritis. The antioxidant activity of ethanolic and water extracts from a Taiwanese musculoskeletal herb, Milk-Fig-Tree (Ficus erecta Thumb. var. beecheyana King) was evalulated by various assays including DPPH free radical and superoxide scavenging assays, lipid peroxidation assay, and hydroxyl radical-induced DNA strand scission assay. In addition, total phenolics in the extracts were determined. The results indicate that neither ethanolic nor water extracts significantly scavenged DPPH radicals (IC(subscript 50) values are 2.65 mg/mL and 3.66 mg/mL, respectively) or superoxide (IC(subscript 50) values are 3.71 mg/mL and >5 mg/mL, respectively); however, both extracts had moresignificant effects than catechin on the inhibition of lipid peroxidation in rat liver (P<0.05). Moreover, both extracts protected ΦXl74 supercoiled DNA against strand scission induced by UV photolysis and H2O2. The total phenolics content of the ethanolic extract was about twice that of the water extract, which is well correlated with the observed results that the ethanolic extract possesses stronger antioxidant activity than the water extract. |
