中文摘要 | 本研究探討不同細胞固定化載體進行ABE醱酵生產丁醇的效率,結果發現固定化於磚頭粒子載體的丁醇生產菌"Clostridium acetobutylicum" CPCNE菌體呈現長桿狀形態且活動力較佳,細胞活性維持在較高狀態,有利菌體生長及進行醱酵作用。依葡萄糖利用效率結果推測磚頭、發泡沸石、海綿、玉米稈及棉為較佳的固定化載體選擇;依丁醇產量結果發現磚頭、海綿、不織布、發泡沸石、玉米稈及棉為較佳的載體選擇。各載體以粒徑2~3.36 mm的磚頭粒子固定化效果最好,於48小時即可產生12.7 g/L丁醇、22.3 g/L總溶劑、轉化率為0.32 g/g、產率為0.46 g/L/hr。經過連續6次批次醱酵實驗,磚頭粒子(粒徑_2~3.36 mm)實驗組皆可醱酵產生>10.8 g/L丁醇,證實磚頭粒子的細胞固定化穩定性佳,適合於長時間連續醱酵操作。相較於其他載體,磚頭粒子的微生物固定化醱酵效果最佳,且磚頭粒子重量較重,在醱酵反應器中易沉降,便於與液態溶劑產物分離,所以最適合作為固定化載體使用。 |
英文摘要 | In this study, the efficiency of butanol production by ABE fermentation in immobilized cell reactors by using different carriers was discussed. The results showed that "Clostridium acetobutylicum" CPCNE immobilized in bricks performed long rod-shaped, high cell activity and better fermentation efficiency. By the assay of glucose-consuming efficiency, it was suggested that the brick, boiling chip, corn stalk and cotton were appropriate carriers for the immobilization of "Clostridium acetobutylicum" CPCNE. By the assay of butanol-producing efficiency, it was suggested that the brick, nonwoven, boiling chip, corn stalk and cotton were better carriers for the immobilization of "Clostridium acetobutylicum" CPCNE. The cells immobilized in brick pieces (2~3.36 mm size) performed the best butanol-producing ability while 12.7 g/L butanol and 22.3 g/L ABE solvent were produced with ABE yield and productivity of 0.32 g/g and 0.46 g/L/h, respectively, following 48 hr of fermentation. In six consecutive batch fermentation experiments, the cells immobilized in brick pieces all performed good butanol-producing efficiency with more than 10.8 g/L butanol, indicating that the cells immobilized in brick pieces were stable and suitable for the long-term continuous fermentation. Compared to other carriers, brick pieces were the most suitable for immobilization of "Clostridium acetobutylicum" CPCNE because they could be easy to sink under the bottom of the bioreactor to separate from solvent produced by ABE fermentation. |
