中文摘要 | 本研究利用植物組織培養微體繁殖技術誘導金門地區原生唐杜鵑("Rhododendron simsii" Planch.)莖節培殖體增殖側芽,將芽體以不同方式在幾種介質中進行植株發根試驗,並馴化小苗以培育綠美化可用之苗木。在芽體增殖試驗中,以34.4及49.2 μM 6-(γ-γ-dimethylallylamino) purine(2ip)處理含頂芽及去除頂芽之培殖體皆能產生較多大芽,添加6-benzylaminopurine(BAP)的處理中,培殖體培養2週(wk)時即出現褐化情形。試管內("invitro")芽體發根試驗顯示,根系發展狀況以紗布為發根介質者,優於以洋菜為介質者。將芽體浸泡在4.92×10^3 μM indole-3-butyric acid(IBA)2分鐘誘導發根效果最佳;試管外("ex vitro")發根試驗中,以水苔為介質者優於其它處理,可獲得較多之根量(7.32 roots perexplant),但於混合介質中者根長較長,達2.54 cm。發根芽體後續馴化成活率亦以混合介質處理者較高,達68.7%,且生長情形較佳。本研究利用微體繁殖方法提供了金門地區唐杜鵑繁殖的技術。 |
英文摘要 | In this study, the shoot tip segments of "Rhododendron simsii" Planch., a native ornamental species in Kinmen, was induced to form multiple axillary shoots through micropropagation technique. Different methods and rooting media were examined to achieve explant rooting for the production of greening plants prepared for planting in the local parks. In the axillary shoot proliferation experiments, the treatment of 34.4 and 49.2 μM 6-(γ-γ- dimethylallylamino) purine (2ip) on shoot tip explants either with or without apical buds obtained better results (more large shoots). However, the 6-benzylaminopurine (BAP) treatment was not suitable for shoot induction and conducted to explant browning within 2 wk in culture. In the "in vitro" rooting experiment using cheesecloth as a replacement of agar medium, the root development was better achieved. A 2-min basal soaking of the shoots in 4.92×10^3 μM indole-3- butyric acid (IBA) obtained the greatest rooting performance. In the "ex vitro" rooting experiment using sphagnum as the acclimatization medium, well-developed roots (7.32 per explant) were greatly obtained as compared to those incubated in other media. Shoots directly acclimatized in a mixed medium (peat moss: vermiculite: perlite, 1: 1: 2 by volume) obtained the best elongation (2.54 cm) and also showed a better survival rate (68.7%). The present study provided a feasible micropropagation procedure to propagate "R. simsii" for planting in Kinmen area. |
