中文摘要 | 微透析取樣技術是生物醫學分析上非常省時、簡便之取樣技術。使用之微透析探針採用商業化產品。樣品溶解及取樣過程僅使用純水。取樣、淨化及分析過程是在直接、動態、自動化中完成。本研究發展之線上微透析取樣技術結合高效液相層析/紫外光偵測系統,可同時定量美白化妝水、乳液、乳膏中維他命C葡萄糖苷、麴酸、維他命B₃。基於真實樣品之分析物不是微量及偵測波長之剛硬性的考量下,本實驗選擇254nm做為後續分析之偵測波長。測得維他命C葡萄糖苷、麴酸、維他命B₃之濃度線性範圍分別介於0.068-304、0.071-284、0.024-488 µg mL¯¹,線性相關係數依序是0.9982、0.9999、0.9999,偵測極限分別為0.01、0.01、0.007 µg mL¯¹。添加回收率介於92~106%,變異係數介於3.9~8.7%。本研究將所開發之分析方法,應用在六種真實樣品中之美白劑含量測定。測得之結果顯示本分析方法具高精密度與高準確度。 |
英文摘要 | Microdialysis sampling (MDS) technique is a simple and easy process and frequently use in biomedical analyses. Microdialysis probe used was a commercial product. Only deionized water was used as the solvent in the sample dissolved and sampling. The sampling, clean-up, and analysis processes can be accomplished in direct, dynamic, and automatic procedures. A simultaneous determination of ascorbyl glucoside (AA-2G), kojic acid (KA), and niacinamide (VitB₃) in bleaching cosmetics using on-line MDS coupled with HPLC/UV has been developed. Considering the rigidness of detection wavelength and the levels of the analytes in the samples are high (0.1%-2%). The analytes were all detected by ultraviolet light absorption at the wavelength of 254 nm. Calibration curves were found to be linear in the 0.068~304 (AA-2G), 0.071~284 (KA), and 0.024~488μg mL¯¹ (VitB₃) respectively. The correlation coefficients of linear regression analysis were with the range 0.9982-0.9999. The detection limits of these compounds are 0.01 for AA-2G, 0.01 for KA, and 0.007 μg mL¯¹ for VitB₃ respectively. The recovery values were in the range 92-106% with good reproducibility (R.S.D. = 3.9-83.7%). Six commercial available bleaching cosmetics were assayed using the procedure developed in this study. The results obtained confirm the precision of the method and confirm the label claim on the cosmetics. |
