中文摘要 | THS2-3菌株是從東海大學校園土壤中分離,有生產蛋白�t潛力的芽孢桿菌。所產生 的蛋白�t受EDTA的抑制,若以鋅離子處理可解除部分抑制的作用。此酵素最佳的催化反應pH 值是在7∼9之間,因此,這是一種以鋅離子為反應中心的金屬型中性蛋白�t。這種酵素並非 耐熱型的thermolysin,但經40℃,30分鐘的熱處理,酵素活性仍然可以保持100%,在Ca�� 離子溶液中耐熱性更能提高。酵素分子量以分子篩方法測得為41,000道耳頓。以酪蛋白為基 質,Km值是2.2mg/ml,Vmax是5.05μg/ml/min。 |
英文摘要 | Bacillus sp. THS2-3, a potential strain for producing protease, was isolated from the soil of Tunghai University. The protease of this strain was showed to be inhibited by EDTA, but Zn�パon is able to recover partial activity from the inhibition. The optimum pH range for this enzyme reaction is 7∼9. It is believed that the enzyme is a metal neutral protease with Zn�パon in the active site. Although this protease is not so thermal stable as thermolysin, it could maintain 100% activity after 30 min incubation at 40℃ water bath. The molecular weight of this protease is about 41,000 daltons determined with Sephadex G-200 molecular sieve chromatography. Using casein as substrate, the Km and Vmax are 2.2mg/ml and 5.05μg/ml/min, respectively. |
