中文摘要 | 目的:台灣牛樟芝(Antrodia camphorate)是一種台灣原生種的食用菌類藥物,根據研究樟芝多醋體萃取物(Antrodia camphorate polysaccharides extracts)之生物效果最佳,近年來有研究指出它具有抗氧他和抗突變功效,然而對乳癌細胞的抑制效果則未曾有系統的研究;台灣的婦女中乳癌是最常見的惡性腫瘤,臨床上乳癌的發生率近年來有顯著上升的趨勢,因此如何預防與治療乳癌已成為重要的課題。方法:本計劃以MCF-7 cell line(human breast cancer cell line;人類乳癌細胞株)進行體外培養,探討樟芝萃取物對於乳癌細胞凋亡(apoptosis)訊息路徑的影響,我們以流式細胞儀(flow cytometry)分析癌細胞之細胞週期、細胞凋亡以及使用西方墨點法分析細胞凋亡相關訊息路徑變他情形與下游蛋白質的表現,以進一步探討樟芝萃取物抑制乳癌細胞之機轉。結果:研究結果顯示,細胞在加不同濃度之樟芝萃取物後,經流式細胞儀(flow cytometry)分析後發現樟芝萃取物明顯誘發細胞凋亡反應,顯示樟芝萃取物確實具有促使癌細胞細胞凋亡之能力,然後經西方墨點法分析後發現,樟芝萃取物亦明顯誘發細胞凋亡蛋白caspase3、caspase7、PARP與Cyt-C等,研究結果顯示樟芝萃取物藉由誘發細胞凋亡蛋白之作用機轉來達成抑制癌細胞之效果。結論:因此本研究成功證實樟芝萃取物具有抗乳癌細胞之能力與其可能之抗癌作用機轉,本研究不止促進抗癌細胞相關機制的暸解,亦可開發另一種抗癌之輔助癌法。 |
英文摘要 | Objective: Antrodia camphorate (Chinese name, niu-chang-chih or chang-chih) is an exclusive fungus parasitic on the inner cavity of the endemic species Cinnamomum kanehirai Hayata. The protective effects of anti-oxidant and anti-mutagenesis of Antrodia camphorate were previous well discovered but the anti-carcinogenicity by Antrodia camphorate polysaccharides extracts against breast cancer were never been systemic evaluated. The purpose of this study is to verify the inhibitory effect on breast cancer cells by Antrodia camphorate polysaccharides extracts. Methods: The evaluation methods including modulation of signal transduction of apoptosis, regulation of cell cycle by flow cytometry, and apoptosis proteins analysis by western blots. Results: The results indicated that the apoptosis of MCF-7 were significantly induced, and the apoptotic protein levels of caspase3, caspase7, parp, cyt-c were dose-related to the Antrodia camphorate polysaccharides extracts. The results of present study can provide the experimental evidence that potential inhibitory effect on anti-carcinogenicity of breast tumors by Antrodia camphorate polysaccharides extracts. Conclusions: The results of present study will prove to be helpful for understanding the anti-carcinogenesis mechanisms by Antrodia camphorate polysaccharides extracts and provide novel insights in controlling breast cancer in our population. |
