中文摘要 | 許多魚類病原菌及多種人類來源鏈球菌的胞外產物(extracellular products, ECP)都已被證實為重要的致病因子。本實驗以魚類來源鏈球菌之ECP進行細胞毒性測試,發現其具有促進細胞增生的能力。故本研究的目標在探究魚類來源鏈球菌ECP中促進細胞增生的特性,及其對體外培養細胞的增生效果。試驗結果顯示,鏈球菌ECP沒有明顯細胞毒性,在稀釋後均有促進吳郭魚卵巢細胞(TO-2 cell)增生的效果,以MTT(3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide)試驗測得的吸光值與對照組比較有顯著差異(P<0.05),細胞增生百分比可達150%;且由試驗可知其ECP可促進細胞DNA合成並改變TO-2細胞的生長曲線。比較不同品種鏈球菌、親水性產氣單胞菌(Aeromonas hydrophila)、愛德華氏菌(Edwardsiella tarda)與溶藻弧菌(Vibrio alginolyticus)之ECP效果,結果發現各鏈球菌菌株如魚類來源鏈球菌(Enterococcus faecalis、Lactococcus garvieae及Streptococcus iniae)、哺乳類動物來源之化膿鏈球菌(Streptococcus pyogenes)及來自底泥之L. lactis,其ECP蛋白質濃度在0.8-4μg/mL時,有顯著性促進細胞增生的能力。其他菌種如A.hydrophila、E tarda與V. alginolyticus的ECP則沒有此能力,其中又以A. hydrophila的ECP具有較強的細胞毒性。 |
英文摘要 | The extracellular products (ECP) were proved to be the virulent factors of many bacterial pathogens. The purpose of this research was to study the cytotoxic activity of ECP(subscript s) from streptococcus and other bacteria to tilapia ovary (TO-2) cell. We compare by MU (3-[4,5-dimet-hylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide) test the cytotoxic effect of ECP derived from different bacteria species including Enterococcus faecalis、Lactococcus garvieae、Streptococcus iniae、Streptococcus pyogenes、Lactococcus lactis、Aeromonas hydrophila、Edwardsiella tarda and Vibrio alginolyticus. The results appeared that the cell proliferation rate were 150% and had obvious difference when streptococcals ECP treated group compare with PBS control group, including Streptococcus spp. Lactococcus spp. and E. faecalis. From the MTT test and the results of 3H-thymidine incorporation test and cell growth curve, it appeared that the ECP from streptococcus group had the mitogenic activity and the ECP from others bacterial pathogens appeared cytotoxic effect especially the Aeromonas hydrophila. |
