中文摘要 | 川芎嗪(Tetramethylpyrazine; TMP)是中藥川芎根莖抽出的有效成分,雖然主要用於人體腦中風和心絞痛的治療,但並未經過嚴密的動物實驗研究。由於被動迴避儀(passive avoidance task)可探討缺血性大鼠學習記憶障礙,而自發運動量可反應動物活動能力,與腦缺血造成肢體活動障礙相關。因此本實驗以上述三者為指標,將大鼠雙側頸動脈與右中腦動脈結紮90分鐘後再灌注,並在術前60分鐘腹腔注射不同濃度的川芎嗪(TMP),用於探討川芎嗪改善大鼠大腦動脈阻塞所致的腦皮質結構損害並且評估改善記憶功能與恢復自發運動的能力。另以大鼠製造腦缺血模型,用免疫化學分析法證明川芎嗪對缺血損傷之腦神經細胞是否具有保護作用。研究結果得知,川芎嗪可以減少腦缺血體積,提高避暗反應時間,改善動物自發運動量。同時發現川芎玳可以抑制神經蛋白被破壞,阻止腦神經損傷,抑制腦細胞凋亡。結論:川芎嗪可以改善腦缺血,對於腦梗塞所造成的缺血性痴呆與行動能力應具有治療作用。川芎嗪也可保護腦神經特定蛋白,阻止缺血腦細胞的發炎,以上研究可供臨床治療腦梗塞中風病人的參考。 |
英文摘要 | Tetramethylpyrazine (TMP) was extracted from Ligusticum chuanxiong, The major indication of TMP were stroke and cardio vascular disease but it never studied on the field of memory and immunochemistry. Owing to passive avoidance task could be used to evaluate the ability of memorizeand autolocalmotor test can response the activity of animal. It was relative to limbs disfunction caused by brain ischemia. The purpose of this study was to detect the above effects of TMP on ischemic/reperfusion rat. Another aim was to detec the function and mechanism of TMP. By immunochemistry methods to prove thatTMP has the effects on protecting injured cerebral vein. The results showed that TMP reduce infarct volume, to prolong the time of step-through latency and to improve autolocomotor activity. The effects of TMP to inhibit neuron protein be destroyed, to reduce iuflammation of neuron and to prevent brain injury were discovered. Conclusion: TMP could improve ischemic condition, to treat the ischemic memory loss and motor activity weakness caused by cranial vein infarction. It also can protect special protein of cerebral neuron and to inhibit inflammation of brain neuron. It is possible to provide this study to clinical treatment of cerebral vein infarcted patient. |
