英文摘要 | Two recent enterovirus 71(EV71) disease outbreaks in Asia resulted in many severe enurologic diseases and rapid death. Because of increased public helath concerns associated with EV71 and its potential for greater enurovirulence, development of a diagnostic method that can rapidly differentiate EV71 from other enterovirus infections has become obligatory. In our study a strategy of multiplex reverse transcription-semi-nested polymerase chain reaction (MRT-SNPCR) was applied to identify enteroviral serotypes in a imple one-tube reaction. The expected amplification products of individual MRT-SNPCRs for enteroviruses were obtained. Primers specific for the enterovirus genus resulted in a 305bp PCR product from all extracts of enteroviral culture supernatants. Further, size-differential PCR products of 158, 185, and 201 bp specific for EV71, coxackivrirus A16 (CAV16), and poliovirus were visualized as predicted, respectively. The assay was demonstrated to be able to detect entreroviral infectionand concomitantly differentiate potential enurovirulent enteroviral serotypes. |
