中文摘要 | 停經後骨質疏鬆症的產生是由於骨生成與骨吸收作用不平衡所造成。停經後,第一型介白質 (interleukin-l, IL-l) 和腫瘤壞死因子 (tumor necrosis factor, TNF) 分泌增加可以刺激IL-6和IL-11的產生,進而刺激骨吸收的進行,造成骨質大量流失或骨質疏鬆的情形。但IL-6對造骨細胞分化有何影響,並沒有定論。因此,本研究旨在利用胎鼠頭蓋骨細胞 (calvarial cell) 體外骨生成模式,探討分級劑量的IL-6對造骨細胞分化的影響。實驗過程中,使用膠原酵素分離法,細胞培養,von Kossa染色法,鹼性磷酸時測定法,氫三放射性標定胸腺嘧啶嵌入法、膠原蛋白合成測定法,及光源鏡檢來分析IL-6對骨生成和骨組織細胞的影響情形。本實驗的結果顯示第六型介白質可減少骨群體的形成和氫三放射性標定胸腺嘧啶的嵌入,進而抑制造骨細胞分化的進行,並且隨著第六型介白質劑量的增加,抑制作用愈明顯。 |
英文摘要 | Osteoporosis is a disease with less bone tissue than normal and predisponse to fracture with minimal trauma. Postmenopausal osteoporosis has been shown to occur as a result of imbalance between bone formation and bone resorption and is closely related to the secretion of interleukin-6 (IL-6) and interleukin一II (IL-11). Howeven, the precise effect of IL -6 on bone cell differentiation is still unclear. The purpose of this study was to test and investigate the effects of IL -6 on osteoblast differentiation using in vitro calvarial cell culture system. The effect of IL -6 was evaluated by bone colony formation assay, alkaline phosphatase activity, 3H-thymidine incorporation assay and collagen synthesis assay. The results of this study indicated that IL -6 exerted a potent dose-dependent inhibition on osteoblast differentiation by reducing the number of bone colonies, 3H-thymidine incorporation and .alkaline phosphatase activity. |
