中文摘要 | 將且B. circulans來源的乳糖醜以嵌入法固定於纖維三醋酸醋(CTA)中。以 50units/gCTA之酵素用量在600rpm之乳化速度下所製得之纖維嵌入化B. circulans乳糖□ ,其活性效率最高。以5%乳糖溶液為基質,在200rpm振盪速率下所測得之活性效率可達 2%左右。B. circulans乳糖□經纖維嵌入固定化後,其酵素特性包括最適作用溫度、pH 及熱穩定性等,均與原酵素相近。由於基質擴散限制(diffusion limitation)之原因,纖 維固定化酵素之Km值比原酵素大10倍,各為115mM及l1.2mM。在50mM半乳糖存在下,纖維 嵌入化B. circulans乳糖□及其原酵素之Ki值則分別為285mM及163mM,而Lineweaver- Burk圖形顯示半乳糖為B. circulans乳糖□之競爭性抑制劑。 |
英文摘要 | Bacillus circulans lactase was entrapped into cellulose triacetate ( CTA) fibers. The optimal conditions for enzyme entrapment were as follows. The ratio of enzyme to CTA was 50 units to 1 g and the emulsifying speed was 600 rpm. When a 5% lactose solution was used as the substrate, at a 200 rpm shaking speed, the activity efficiency could reach 32%. Enzyme properties, such as the optimal temperature, optimal pH and thermal stability of cellulose triacetate entrapped B. circulans lactose, were similar to those of the enzyme's native form. Due to the diffusion limitation for the substrate, the Km of the fiber entrapped enzyme was about 1 0 times that of its soluble form, i.e., 115 mM vs 11.2 mM, when lactose was used as the substrate. In the presence of 50 mM galactose, the Ki of the native and immobilized B. circulans lactase were 163 mM and 285 mM, respectively. In addition, the Lineweaver-Burk plot shows that galactose is the competitive inhibitor for lactase. |
