英文摘要 | Rice sucrose synthase (RSuS) isozymes are encoded by three isogenes, RSus1, RSus2 and RSus3. RSus cDNAs were introduced into E. coli for expression using the T7 promoter/RNA polymerase system. Only the production of recombinant RSuS1 in a homotetrameric form was achieved. The expression of RSus2 and RSus3 cDNA in E. coli resulted in truncated expression products and the accumulation of inclusion bodies, respectively. The recombinant RSuS1 homotetramer was purified from E. coli extract using ammonium sulfate precipitation, Sepharose CL-6B gel filtration and DEAE-Sephacel chromatographies. The enzyme possesses similar biochemical properties as the plant SuS. By comparing the electrophoretic mobilities of RSuS isozymes produced in E. coli and those from various rice tissues, it revealed that the expression site of RSus3 is grain-specific and the expression products of RSus1 and RSus2 are widely distributed in various rice tissues. |
