中文摘要 | 本研究選用16頭9~10月齡藍瑞斯新母豬,供探討子宮內灌注生理劑量的17β-雌 素二醇(17β-E□)與/或前列腺素E□(PGE□)對維持其黃體功能之影響。新母豬於動 情週期Day7前(發情第一日為Day 0),先裝置兩側子宮角導管與一側頸靜脈導管,再將 新母豬逢機分成4組(每組4頭)'分別進行子宮內灌注佐劑、17β-E□、PGE□與17β-E□ +PGE□。17β-E□(3μg/每日)於Days 11~15期間,而PGE□(2.4mg/每日)則於Day 7至Day 22或至發情之日,將每日劑量分6次處理,每次灌注兩側子宮角。並於Days 7~27 每日分兩次採血,供血漿孕酮(progesterone)濃度分析。各組新母豬處理時平均動情週 期日數與處理前者間並無差異,且各組新母豬於處理時平均動情週期日數間,亦未呈差異 ;由此顯示新母豬經子宮內灌注17β-E□與/或PGE□未能延長其動情週期日數。在Days 7~12以及Days18~27各日,各組平均血漿孕酮濃度間均未呈差異。不過,17β-E□組平均 孕酮濃度自週期Day 13(17.7 vs 13.0 ng/mL)、Day 14(13.8 vs 9.0 ng/mL)至 Day 15(10.4 vs 3.6 ng/mL),而PGE□組孕酮濃度則於Day 15(9.3 vs 3.6 ng/mL)均 分別較佐劑組者高(P<0.05);惟17β-E□十PGE鑾組新母豬平均孕酮濃度變化圖象則與 佐劑組者相仿# 330 1$a。綜前提示,在本試驗所使用的劑量與頻率之情況下,單獨17β -E□或PGE□在新母豬黃體解體階段,各具延遲血漿孕酮濃度下降3日或1日之功能,惟兩 者在維持黃體功能中則未具協同作用。 A total of sixteen Landrace gilts, 9 to 10 months of age, was used to study the effects of 17β -estradiol (17β-E□) and prostaglandin E□ (PGE□) on the maintenance of luteal function. Cannulae were surgically inserted into the lumen of each uterine horn for hormone infusion and into jugular vein for blood collection before Day 7 postestrus. Gilts were randomly assigned to one of the 4 treatment groups (4/group) and given daily intrauterine infusions of vehicle, 17β-E□, PGE□, or 17β-E□+PGE□ to both horns, respectively. 17β-E□ (3μg/day) infusions were initiated on Day 11 every 4 hr till Day 15, while PGE□ (2.4 mg/day) infusions were given on Day 7 every 4 hr till estrus or Day 22. Blood samples were collected twice daily from Days 7 to 27 for progesterone (P□) analysis. The results showed that the treatment cycle length did not differ from the previous cycle length in each group in the gilts. In addition, there were no differences in the estrous cycle length between groups of the four treatments, indicating that intrauterine infusions of 17β-E□ and/or PGE□ were ineffective in prolonging interestrous intervals in the gilts. The mean plasma P4 concentrations for 17β-E□ and/or PGE□ treatment groups were not different from the vehicle group on Days 7~12 or Days 18~27 of the cycle. However, the mean plasma P□ concentration of 17β-E□ group was greater (P<0.05) than that of vehicle group from Day 13 (17.7 vs 13.0 ng/mL), Day 14 (13.8 vs 9.0 ng/mL) to Day 15 (10.4 vs 3.6 ng/mL), while the mean plasma P□ concentration for the PGE□ group (9.3 vs 3.6 ng/mL) was greater (P< 0.05) than the vehicle group on Day 15 of the estrus cycle. In contrast, the 17β-E□+PGE□ group had the same profile of P□ level as the vehicle group during Days 7~20 of the estrous cycle. Therefore, it is concluded that both 17β-E□ and PGE□ treatment alone may retard the decline Of P□ secretion for three and one days more, respectively, by the corpus luteum, but the combination treatment do not act synergistically to maintain luteal function during early pregnancy in gilts. |
