中文摘要 | 蕨藻是臺灣沿海地區普遍生長的綠藻,雖早有報告敘述其生長區域,但是對於其藻 種遺傳資訊的描述則闕如。本論文即選取臺灣沿岸地區不同地域的蕨藻種類,先依形態上區 別,再分別由各植株抽取其genomic DNA,以逢機多型性分析指紋圖譜,解析遺傳特性。 逢機多型性是由二十個蕨藻採樣個體的genomic DNA增幅大,共使用十五個10-mer引 子(primer),其中有十四個產生增幅條帶,由增幅條帶的有或無所換算得到的相似性係數,二 十個蕨藻採樣由0.35至0.91,在採得的四種變種(C. racemosa var. microphysa, macrophysa, peltata and laetevirens)中,其相似性係數並沒有比種間之比值(和C. scalpelliformis比較)及 其它藻類較大,依逢機多型性的遺傳形質分析,此四變種應定為分別的不同種。 二十個蕨藻個體採樣共有兩種、四變種等不同區域族群,其親緣性樹狀圖顯示,不同區 域地理族群,其遺傳特性並沒受黑潮的影響而有方向性區域的差異。臺灣沿海地區的蕨藻其 相似性係數、親緣性樹狀圖顯示其種間、種內的差異並無明顯地理分布位置上的區隔。 |
英文摘要 | Caulerpa species are common taxa of green algae in northern and southern Taiwan seashore. The distribution of Caulerpa species in Taiwan has been documented, however there is no further genetic data for geographic Caulerpa species in Taiwan. In this study, Caulerpa specimens were collected around Taiwan Island and distinguished by morphological characteristics. Genetic similarity was then estimated using the RAPD (random amplified polymorphic DNA) markers. RAPD markers were amplified from genomic DNA of 20 collections. Fifteen oligonucleotide 10-mer were used as primers to generate DNA profiles for each individual. Fourteen of the fifteen primers were positive and band-producing. The similarity was calculated from a C. scalpelliformis and C. racemosa with four varieties of geographic Caulerpa. The similarity was varied from 0.35 to 0.91 among the collected samples. The similarity among C. racemosa var. microphysa, macrophysa, peltata and laetevirens for the study were not larger than that with C. scalpelliformis. It is suggested that C. racemosa var. microphysa, macrophysa, peltata and laetevirens should belong to different species in Caulerpa genus basing on this RAPD analysis. The dendrogram shows the relationship of the 20 collections including two species and four varieties of Caulerpa populations and reveals that variations among geographic specimens are not directed by the Kuroshio Current. The difference in genetic characteristics among Taiwan Caulerpa species does not correlated to directional geographic position. The inter-specific and intra-specific variation and phylogenetic dendrogram among geographic Caulerpa Specimens in Taiwan show no significant difference. |
