中文摘要 | 本研究主要目的為探討經氣管灌注途徑重覆暴露鎘之肺毒性傷害,以氣管套針經 由氣管內快速注入法直接投予 0、0.3、3 及 30ug/kg 不同劑量之氯化鎘連續 4 週, 每週 5 天。 結果顯示,高劑量組 (30ug/kg) 於處理後第 3 週雌鼠體重逐漸減輕,第 4 週各處 理組之雌雄鼠增重均較對照組顯著下降, 並呈現藥劑劑量與增重負相關性;30ug/kg 處理組 肺臟重量增加及腎臟重量減輕,0.3 及 3ug/kg 組之臟器重量均無顯著變化; 各組血液相亦 無差異,血清肌酐酸磷酸脢值則於各處理組均上升; 肺臟、肝臟、腎臟及血液中均呈劑量與 殘留量正相關性,其中以肺臟最為顯著。肉眼病變可見大鼠肺臟腫脹,表面有多發性局部灰 白色病變,光以高劑量組最為明顯。肺臟顯微病變之終末支氣管部分上皮脫落壞死,上皮細 胞呈現立方狀及多層性增生,且排列紊亂突出管腔內,病變區肺巨噬細胞及嗜中性球浸潤, 3 及 30ug/kg 組大鼠肺臟有明顯局部肉芽腫及纖維化病變。 由本實驗結果顯示,以氣管內 快速注入法長期暴露氯化鎘可造成肺臟明顯傷害,其對大鼠之無毒害作用劑量 (noobserved effect level, NOEL) 為 0.3ug/kg( ≒ 2.8ug/m �� ), 而最低毒害作用劑量 (minimal toxic level, MTL) 為 3ug/kg( ≒ 28ug/m �� )o |
英文摘要 | The pulmonary toxicity of cadmium chloride was investigated by repeated intratracheal instillation in rats. "The rats were dosed with cadmium chloride at 0, 0.3, 3, and 30 ug/kg body weight, respectively, and continuously treated 5 day/week for 4 weeks. The results revealed that the body weight gain of females decreased at third week in the 30 ug/kg group. The body weight gain of treated groups decreased at the end of treatment. This tendency showed negative dose-response relationship. Either increase lung or decrease kidney weight was noted in the 30 ug/kg group,but there were no effect in the 0.3 and 3 ug/kg groups. No significant changes of hematological results were observed, but enzymatic activity of CPK was significantly increased in the treated rats. The cadmium concentration showed a dose-related increase in the lung, liver, kidney and whole blood, and increased obviously in lung tissue. At necropsy, enlargement, and multifocal grayish foci were found in the affected lungs, and the 30 ug/kg/bw rats presented most severity among the treated groups. Microscopically, the desquamated epithelia, proliferated bronchiolar epithelia arranged in an irregular pattern, and protruded into the lumen cavity were observed in the terminal bronchioles. Enlargement of nuclei, multiple cuboidal cell hyperplasia, numerous macrophages and neutrophils infiltrated in the affected areas. Focal granulomatous and fibrotic lesions of lungs were also presented at the doses of 3 and 30 ug/kg treated-rats. In conclusion, cadmium chloride can induce pulmonary toxicity after intratracheal exposure in rats, and the no observed effect level (NOEL) and the minimal toxic level (MTL) are 0.3 @g/kg (=2.8 ug/m �� ) and 3 ug/kg (@28 ug/m �� ) in this study trail, respectively. |
