中文摘要 | 以臺灣雲杉(Picea morrisonicola Hayata)成熟胚為培植體進行培養,誘導癒合組織及胚原性癒合組織。結果顯示以MS為基礎培養基培養效果較佳,而添加1ppm BA及 NAA時胚的成活率最高。在黑暗下以添加1 ppm 2,4-D與1 ppm BA時癒合組織的誘導率最高。在全光照環境中培養,可誘導出綠色、緊實之癒合組織,而在黑暗下培養則誘導出黃色、鬆軟之癒合組織。將黃色、鬆軟癒合組織培養於添加2 ppm 2,4-D及1 ppm BA的MS培養基中,在黑暗環境下培養四個月,可誘導出白色黏質具有胚性端及懸柄細胞的胚原性癒合組織;將該胚原性癒合組織移至添加0.1 ppm 2,4-D、1 ppm ABA的MS固態培養基中,經過兩個月全光照的培養,則可誘導出綠色前子葉期體胚。 |
英文摘要 | Mature embryos of Picea morrisonicola Hayata were used for callus and embryogenic callus induction. Murashige and Skoog (MS) medium was found to be a better culture medium compared to WPM and B5 Embryos had higher survival rate on MS medium supplemented with 1 ppm NAA (α-naphthaleneacetic acid). Under dark condition, higher callus induction rate was obtained on MS medium supplemented with 1 ppm 2,4-D (2, 4-dichlorophenoxyacetic acid) and 1 ppm BA (6-benzylaminopurine). Light condition affected the nature of the callus. Green compact and yellowish soft callus were formed when cultures were incubated under light and dark condition, respectively. When yellowish soft callus were transferred to MS medium with 2 ppm 2,4-D and 1 ppm BA, and incubated under dark condition for 4 months, white and sticky embryogenic callus with numerous somatic embryos having embryonal head and suspensor were formed. Green cotyledonary stage somatic embryos were developed after embryogenic callus were transferred to MS solid medium supplemented with 1 ppm 2,4-D 1 ppm NAA and 2 ppm ABA and incubated under light condition for 2 months. |
