中文摘要 | 以臺灣栽培之洛神葵(Hibiscus sabdariffa L.)為參試材料,觀察根尖細包有絲分裂(mitosis)染色體數目與花粉母細胞第一次減數分列(meiosis I)染色體配對情形,並配合體細胞染色體核型分析(karyotype analysis),以瞭解染色體組(genome)組成,確定其倍數性(polyploidy)。 根尖細胞染色體數目證實為2n=72,花粉母細胞減數分裂染色體對數為24,配合核型分析,推測洛神葵應為2n=6x=72之異質六元體(AAABB hexaploid),其結實良好,此結果可直接提供育種雜交實務參考。進一步可與同屬近緣種、野生種雜交,以瞭解其親本來源及種屬間親緣關係。 |
英文摘要 | Hibiscus sabdariffa L. has been cultivated in Taiwan for many years, which is selected as the studied material in this study. An attempt is made to observe the chromosome number in root tip cells during mitotic stage and pollen mother cells during meiotic stage, respectively. In this study, cerotype analysis of somatic chromosome, combined with meiotic study, were used to investigate the genome constitution and the type of polyploidy. The somatic chromosome numbers of the root tip cells were confirmed to be 2n=72, and the numbers in meiotic stage of pollen mother cells were 24. Combining with the karyotype analysis, the chromosome numbers reveal that Hibiscus sabdariffa L. might to be AAABB hexaploid of 2n=6x=72, which had good seed set. The present data have provided some information for breeding. However, the further studies of interspecific and intraspecific hybridization can be very helpful in tracing the material parents of polyploidy and understanding the degree of relatedness. |
